The Kinase Enzyme Systems include a recombinant kinase enzyme, a substrate appropriate for the enzyme, a reaction buffer and supplemental reagents as needed. The p70S6Kb Kinase Enzyme System contains:
- p70S6Kb Kinase, 10μg (Human, recombinant full-length). MW: ~85kDa.
- RSK Substrate (KRRRLSSLRA); derived from human 40S ribosomal protein S6 (amino acids 230–239).
- Kinase Assay Buffer I, DTT.
Recombinant full-length human p70S6Kb was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag. p70S6Kb is a member of the RSK (ribosomal S6 kinase) family of serine/threonine kinases and is activated by mitogenic stimuli, including growth factors, cytokines and phorbol esters. p70S6Kb contains 2 nonidentical kinase catalytic domains and phosphorylates the S6 ribosomal protein and eukaryotic translation initiation factor 4B. Phosphorylation of S6 leads to an increase in protein synthesis and cell proliferation. PI3 kinase pathway and mTOR are involved in the activation of p70S6Kb, but other pathways can also activate this target protein.

Use with ADP-Glo™ Kinase Assay
The ADP-Glo™ Kinase Assay is a luminescent kinase assay that measures ADP formed from a kinase reaction; ADP is converted into ATP, which is a substrate in a reaction catalyzed by Ultra-Glo™ Luciferase that produces light. The luminescent signal positively correlates with ADP amount and kinase activity. The assay is well suited for measuring the effects of chemical compounds on the activity of a broad range of purified kinases, making it ideal for both primary screening as well as kinase selectivity profiling. The ADP-Glo™ Kinase Assay can be used to monitor the activity of virtually any ADP-generating enzyme (e.g., kinase or ATPase) using up to 1mM ATP.
