Efficient Detection of Malate in Diverse Samples

The Malate-Glo™ Assay is a fast and sensitive method for detecting L-malate from various biological samples, such as cultured cells, tissue homogenates and serum. Based on bioluminescent technology, the Malate-Glo™ Assay can detect subtle changes in mitochondrial metabolism and the TCA cycle. Its streamlined sample preparation protocol involves acid treatment and neutralization directly in the assay wells, eliminating the need for cell collection, centrifugation and spin columns. The assay is adaptable to a 384-well format, facilitating quick analysis of many samples.

How the Malate-Glo™ Assay Works

Malate Dehydrogenase catalyzes the oxidation of malate with concomitant reduction of NAD+ to NADH. In the presence of NADH, Reductase enzymatically reduces the proluciferin Reductase Substrate to luciferin. Luciferin is detected using Ultra-Glo™ Recombinant Luciferase, and the amount of light produced is proportional to the amount of malate in the sample.
