Flexible and Efficient Purification of DNA and RNA from FFPE Samples

The Maxwell® RSC XtractAll FFPE DNA/RNA Kit, used with the Maxwell® RSC or Maxwell® RSC 48 Instrument, is an innovative solution designed to efficiently purify DNA and RNA from formalin-fixed, paraffin-embedded (FFPE) tissue samples. This kit streamlines nucleic acid extraction with unparalleled flexibility by offering four distinct workflows allowing users the choice to extract: DNA only, RNA only, total nucleic acid in a single eluate, or sequentially extract DNA and RNA into separate eluates—all from the same sample. This versatility ensures that laboratories can maximize the utility of limited FFPE tissue inputs. The Maxwell® RSC XtractAll FFPE DNA/RNA Kit combines minimal hands-on time and easy-to-follow protocols, with the consistency and efficiency of automated extraction. The Maxwell® RSC XtractAll FFPE DNA/RNA Kit produces high-quality nucleic acids that are suitable for advanced downstream analysis assays, such as digital PCR and next-generation sequencing (NGS).

The Maxwell® RSC Instrument can process 1 to 16 samples, and the Maxwell® RSC 48 Instrument can process 1 to 48 samples, in a single run. The Maxwell® Instruments are supplied with preprogrammed purification procedures and are designed for use with the predispensed reagent cartridges provided in the Maxwell® RSC XtractAll FFPE DNA/RNA Kit.

Note: The Maxwell® RSC XtractAll FFPE DNA/RNA Kit is only compatible with Maxwell® RSC or Maxwell® CSC (RUO mode) Instrument software version 4.0.0 or greater; and Maxwell® RSC 48 or Maxwell® CSC 48 (RUO mode) Instrument software version 4.1.0 or greater.

Maxwell® RSC XtractAll FFPE DNA/RNA Kit workflows. After preparing the FFPE tissue sample lysate, choose to extract DNA, RNA, total nucleic acid, or sequentially extract DNA and RNA. Only one cartridge per sample is needed regardless of the workflow you choose. This kit maximizes flexibility and utilization of limited FFPE samples, while minimizing waste.

Fast and Versatile FFPE DNA and RNA—Without Compromises!

The Maxwell® RSC XtractAll FFPE DNA/RNA Kit significantly reduces sample extraction turnaround time with workflows of ~180 minutes or less, depending on the selected protocol. With this kit, FFPE tissue sample lysates are prepared in approximately 90 minutes, and then the remaining extraction steps are automated by the Maxwell® Instrument for fast, consistent and efficient extraction in only 30–60 minutes. This streamlined approach enables laboratories to reliably and quickly process samples, freeing up valuable time for other laboratory needs.

The Maxwell® RSC XtractAll FFPE DNA/RNA Kit delivers unmatched flexibility with four workflow options. From one FFPE input sample, choose to extract: DNA only, RNA only, total nucleic acid in a single eluate, or sequential DNA and RNA extraction into separate eluates. This adaptability supports a diversity of applications without compromising on yield or quality of results, eliminating the need to perform multiple extractions and benefiting laboratories with limited FFPE sample material.

The Maxwell® RSC XtractAll FFPE DNA/RNA Kit provides consistent yield of DNA and RNA in each workflow. Lung tumor FFPE (Panel A) and colon normal FFPE (Panel B) samples were extracted using the Maxwell® RSC XtractAll FFPE DNA/RNA Kit following the DNA (“DNA”), RNA (“RNA”), total nucleic acid (“TNA”) or sequential DNA and RNA (“Seq DNA” and “Seq RNA”) protocols. The yield of DNA and RNA was evaluated via amplification assays (102bp target for DNA, 100bp target for RNA). Data represent mean ±SD, N=6.

Comparison of DNA and RNA yields from a variety of FFPE tissue samples. DNA and RNA were extracted from multiple types of FFPE tissue samples (breast, liver, colon, kidney, uterine, endometrial, lung, cervical) using the Maxwell® RSC XtractAll FFPE DNA/RNA Kit sequential DNA/RNA workflow and two other commercially available sequential FFPE DNA/RNA extraction kits. DNA yield was evaluated by amplification using the ProNex® DNA QC Assay (Cat.# NG1002) for short (75bp; Panel A) and long (300bp; Panel B) target amplicons. RNA was evaluated by amplification assay for short (100bp; Panel C) and long (171bp; Panel D) target amplicons. The Maxwell® RSC XtractAll FFPE DNA/RNA Kit shows equivalent or superior performance for both DNA and RNA yields when evaluated by amplification assay, especially for longer target amplicons. Performance of the Maxwell® RSC XtractAll FFPE DNA/RNA Kit in digital PCR. Reliable Fluorescent Quantitation for Confident Downstream Assay Performance

Fluorescence dye-based quantitation assays, such as the QuantiFluor® ONE dsDNA System (Cat.# E4871) and QuantiFluor® RNA System (Cat.# E3310), are often used to quickly and conveniently assess the yield of extracted DNA and RNA. In contrast to other FFPE extraction offerings, DNA and RNA extracted with the Maxwell® RSC XtractAll FFPE DNA/RNA Kit produces fluorescence dye quantitation results that more reliably predict success of the purified nucleic acids in amplification assays, such as qPCR and digital PCR. This improved concordance between fluorescence dye quantitation and amplification assays minimizes the risk of failed downstream assays, saving you both time and money.

Assessment of fluorescence- and amplification-based quantitation assay concordance for DNA and RNA extracted from FFPE tissue. DNA and RNA were extracted from eight different FFPE tissue samples using the Maxwell® RSC XtractAll FFPE DNA/RNA Kit and two other commercially available sequential FFPE DNA/RNA extraction kits. Panel A. DNA was quantified using fluorescence-based (QuantiFluor® ONE dsDNA System [Cat.# E4871]) and amplification-based (qPCR; 102bp amplicon) assays. Panel B. RNA was similarly quantified using fluorescence-based (QuantiFluor® RNA System [Cat.# E3310]) and amplification-based (RT-qPCR, 100bp amplicon) assays. The ratio of fluorescence- to amplification-based quantification was calculated and compared (“Fluorescence/Amp Ratio”). A ratio value approaching 1 indicates good concordance of the results between the fluorescence-based and amplification-based quantitation assays. In contrast to the other extraction kits, the Maxwell® RSC XtractAll FFPE DNA/RNA Kit demonstrated fluorescence-based quantitation readouts that more consistently predict DNA and RNA quantitation via amplification-based assays.
