Interpreting Multiplexing Data Using The CellTox™ Green Cytotoxicity Assay

CellTox™ Green Cytotoxicity Assay

The CellTox™ Green Cytotoxicity Assay measures changes in membrane integrity that occur when cells die, and is used to assess cytotoxicity in cell cultures after experimental manipulation. The CellTox™ Green Dye binds to DNA when membrane integrity is compromised, but is excluded from viable cells. When the dye binds DNA, its fluorescence properties are enhanced, and the fluorescent signal is proportional to cytotoxicity. The assay can be multiplexed with other spectrally distinct measures of cell health to provide information on why and how cells are dying. This guide will help you interpret the results from these types of experiments.

Questions About Interpreting Multiplexing Data

1

How do I detect cytotoxicity as it occurs in real-time? How do I measure cytotoxicity over a long time course?

The CellTox™ Green Cytotoxicity Assay can be used to measure cytotoxicity over a long time course and/or to detect cytotoxicity as it occurs in real-time.

2

When should I measure for caspase activation?

Kinetic monitoring of cytotoxicity with the CellTox™ Green Dye can allow you to identify the appropriate point to measure for mechanism of action.

3

My cells are dying! How can I tell if it is apoptosis or necrosis?

Multiplexing cell health assays reveals information about a compound's mechanism of action to help you determine if cells are dying from apoptosis or necrosis.

4

My cells are not proliferating; how do I differentiate nonproliferation from cell death? Should I measure cytotoxicity or cell viability?

Multiplexing cell health assays can be used to distinguish between antiproliferative and cytotoxic effects.
