Module 2 - Cytotoxicity Kinetics and Mechanism of Action
When should I measure for caspase activation?
Correlating Cytotoxicity Kinetics to Mechanism of Action
Kinetic monitoring of cytotoxicity with CellTox™ Green Dye allows users to identify an exact point in time when changes in membrane integrity begin. This can be beneficial for mode-of-action studies where additional biomarkers may be of interest.
The CellTox™ Green Cytotoxicity Assay can be multiplexed with other spectrally distinct measures of cell health to provide mechanistic information relating to cytotoxicity. The detection of caspase-3/7 activation is frequently used as a positive marker for apoptosis. The Caspase-Glo® 3/7 Assay provides an extremely sensitive means of monitoring caspase activation in cell assays.

Bortezomib has been shown to inhibit proteasomes, cellular enzymes that play a role in regulation and cell growth processes, which in turn can lead to cancer cell death. Cells were treated with Bortezomib. The cell response was evaluated over the course of 72 hours using the CellTox™ Green Cytotoxicity Assay.
