GTPase Assays: New Tools to the Rescue
- GTPase signaling overview
- GTPase families and involvement in disease
- A new approach to measure GTPase activity
- Data measuring the effects of GAP and GEF proteins on GTPases
Summary
GTPase proteins are key cell signaling nodes regulating diverse cellular functions. Clinical manifestations associated with deregulation of GTPases, such as seen in XXXX and YYYY, illustrate the importance of this class of hydrolase enzymes. In fact mutations in Ras GTPases are associated with one-third of all human cancers.. However, measuring the activity of these important proteins is difficult. GTPases have a very high affinity for guanine nucleotides and are slow acting enzymes, two factors that make these enzymes difficult to measure. Adding to the complexity is the need for associated proteins to help facilitate GTP hydrolysis and nucleotide exchange.

Subhanjan Mondal is a Senior Research Scientist at Promega. He has been with Promega since 2012. Prior to joining Promega he was a Postdoctoral Fellow at Children’s Hospital Boston/Harvard Medical School. Subhanjan received his Ph.D. in Biochemistry and Cell Biology from the University of Cologne, Germany. His graduate work was in fact on GTPases and he wished that convenient assays for GTPases like the one he developed existed back then.
