Recombinant human CAMK2γ (C-terminal truncation) was expressed by baculovirus in Sf9 cells using an N-terminal GST tag. CAMK2γ is a member of the CAMKII family, which are ubiquitous serine/threonine protein kinases that have been implicated in diverse effects of hormones and neurotransmitters. CAMK2γ has six alternatively spliced variants that encode six different isoforms. Some of these variants have been identified in human tumors. Transgenic mice expressing a partially calcium-independent mutant form of CAMKG showed 1.5- to 2-fold increase in the thymus of these mice, at least in part due to an increase in the life span of double-positive thymocytes. There was an increase in the number of T cells in the secondary lymphoid organs that had acquired an antigen-dependent memory phenotype.

Use with ADP-Glo™ Kinase Assay
The ADP-Glo™ Kinase Assay is a luminescent kinase assay that measures ADP formed from a kinase reaction; ADP is converted into ATP, which is a substrate in a reaction catalyzed by Ultra-Glo™ Luciferase that produces light. The luminescent signal positively correlates with ADP amount and kinase activity. The assay is well suited for measuring the effects of chemical compounds on the activity of a broad range of purified kinases, making it ideal for both primary screening as well as kinase selectivity profiling. The ADP-Glo™ Kinase Assay can be used to monitor the activity of virtually any ADP-generating enzyme (e.g., kinase or ATPase) using up to 1mM ATP.
