Profile Compound Selectivity Against 192 Kinases in Live Cells

The NanoBRET® Target Engagement (TE) K192 Kinase Selectivity System can quantitatively measure intracellular compound occupancy against a panel of 192 kinases in a single experiment. The system relies on the NanoBRET® TE technology, a bioluminescence resonance energy transfer (BRET) method that uses the small, extremely bright NanoLuc® luciferase as the energy donor and a cell-permeable NanoBRET® TE Tracer as the energy acceptor. The K192 Kinase Selectivity System is a live-cell method using an addition-only, multiwell plate format with a single NanoBRET® TE Kinase Assay. The NanoBRET® TE K192 Kinase Selectivity System is optimized for each kinase to provide quantitative occupancy data, enabling cellular profiling of compound-kinase interactions.

NanoBRET® Target Engagement Assay Principle

The NanoBRET® TE K192 Kinase Selectivity System uses the NanoBRET® TE technology, a bioluminescence resonance energy transfer (BRET) method. In this method, a target is expressed in mammalian cells as a target-NanoLuc® fusion protein. BRET is achieved by luminescent energy transfer from NanoLuc® luciferase to a cell-permeable NanoBRET® TE Tracer that is bound to the target-NanoLuc® fusion protein. A test compound that is cell-permeable and competes with the tracer for target binding will result in a loss of NanoBRET® signal. Using the optimized assay conditions provided for each target, the assay yields quantitative intracellular compound affinity and occupancy. Compound occupancy is quantified using controls that allow the BRET signal to be normalized.

K192 Kinase Selectivity System Provides Turn-Key Cellular Profiling

The NanoBRET® TE K192 Kinase Selectivity System contains a NanoBRET® TE K192 Kinase Vector Panel and a single NanoBRET® TE Intracellular Kinase Assay (K-10). The optimized protocol offers an addition-only workflow using multiwell plates. Begin by simply transfecting the vector panel into HEK293 cells or TransfectNow™ HEK293 Cells in a multiwell plate. Following overnight incubation, add your test compounds to the assay plate and perform the NanoBRET® TE Kinase Assay (K-10), according to the optimized protocol provided. A mutimode plate reader is used to measure BRET.

Get The Live-Cell Kinase Selectivity Profiling Data You Need

Cell-free biochemical techniques for measuring binding or activity of kinase inhibitors can fail to accurately predict engagement of kinases inside a cell.

The dendrograms compare the target occupancy obtained with 1µM crizotinib using the live-cell NanoBRET® TE K192 Kinase Selectivity System versus a biochemical cell-free approach. The results showed an improved spectrum of activity in cells, with only 16 targets engaged by crizotinib at 50% occupancy or greater using the NanoBRET® TE method, compared to 49 kinases engaged using the biochemical approach.

The 16 kinases that bound with >50% occupancy using the K192 Kinase Selectivity System are tabularized along with the mean occupancy obtained from three independent experiments. The standard deviations for these measurements indicates that the K192 Kinase Selectivity System is reproducible.

(1a) use Nano-Glo®-branded luminescent assay reagents (LARs) for all determinations of luminescence activity of this product and its derivatives; or
(1b) contact Promega to obtain a license for use of the product and its derivatives with LARs not manufactured by Promega.

For uses of Nano-Glo®-branded LARs intended for energy transfer (such as bioluminescence resonance energy transfer) to acceptors other than a genetically encoded autofluorescent protein, researchers must:
(2a) use NanoBRET®-branded energy acceptors (e.g., BRET-optimized HaloTag® ligands) for all determinations of energy transfer activity by this product and its derivatives; or
(2b) contact Promega to obtain a license for use of the product and its derivatives for energy transfer assays to energy acceptors not manufactured by Promega.

Researchers may transfer derivatives to others for research use provided that at the time of transfer a copy of this label license is given to the recipients and recipients agree to be bound by the terms of this label license. BY USE OF THIS PRODUCT, USER AGREES TO BE BOUND BY THE TERMS OF THIS LIMITED USE LABEL LICENSE. If user is not willing to accept the terms of this label license, and the product is unused, Promega will accept return of the unused product and provide user with a full refund.

User may use this product for research use only; however, no amplification or isolation of the vector components of this product and no transfer or commercial use of this product is allowed. Commercial use means any and all uses of this product by a party in exchange for consideration, including, but not limited to: (1) use in further product manufacture; and (2) resale of the product, whether or not such product is resold for use in research. User shall have no right to manufacture or have a third party manufacture the vector components within this product for any purpose. User shall have no right to modify or otherwise create variations of the nucleotide sequence of the luciferase gene. Notwithstanding the foregoing, user may use this product in provision of services to third parties in exchange for consideration; however, user may not transfer the product to third parties.
