The Wizard® PureWater Kit, compatible with standard laboratory equipment, is specifically engineered for the extraction and purification of DNA from a broad range of water samples, such as water from cooling towers, potable water, and hot- and cold-water systems. This kit is essential for the accurate identification and quantification of bacterial pathogens in water, using PCR, NGS and other molecular techniques. The Wizard® PureWater Kit can handle both concentrated samples collected on a filter and unconcentrated suspension samples, providing flexibility and efficiency in water-quality testing workflows.
The Wizard® PureWater Kit protocol includes three main steps:
- Sample Preparation: Collect and prepare water samples using your currently approved methods, adjusting treatment based on whether samples are concentrated samples collected on a filter or unconcentrated suspension samples to minimize osmotic shock.
- DNA Purification: Mix water samples with lysis buffer and isopropanol, load into a spin column and perform sequential washing steps.
- Elution and Storage: Elute the purified DNA, and store it under recommended conditions until analysis.
This streamlined process ensures the extraction of high-quality DNA that is ready for immediate use in molecular detection tests.
The Wizard® PureWater Kit includes all necessary reagents for 50 DNA isolations, designed to maximize laboratory efficiency and throughput. The kit components are formulated to enhance DNA recovery and purity, which is essential for reliable detection and monitoring of waterborne diseases.

The Wizard® PureWater Kit consistently extracts Gram-positive bacterial DNA from different types of water samples. DNA was extracted from 104CFU/ml live Listeria innocua in 200µl suspension samples (Panel A) or 103CFU/ml live Listeria innocua in 100ml of PBS that was collected onto a filter (Panel B) and purified using the Wizard® PureWater Kit. DNA eluates were assessed using GoTaq® qPCR Master Mix (Cat.# A6001) with primers specific for Listeria. Data represent purification replicates (N = 3); error bars represent standard deviation.
