The SV 96 Total RNA Isolation System provides a high-throughput technique to prepare intact RNA from tissue and cultured cells. Total RNA can be purified from 96 samples in less than an hour without centrifugation. The system incorporates a DNase treatment step that is designed to substantially reduce genomic DNA contamination, which can interfere with amplification-based methodologies. Purification is achieved without phenol:chloroform extraction or ethanol precipitation, and there is no detectable DNase carryover in the final RNA preparation.
Protocols are available for Beckman Coulter and PerkinElmer instruments.

The SV 96 Total RNA Isolation System requires a vacuum pump capable of generating 15–20 inches of Hg or equivalent and the Vac-Man® 96 Vacuum Manifold (Cat.# A2291; see figures) or similar manifold. The Vac-Man® 96 Vacuum Manifold requires the use of an external vacuum trap (not provided).
For automated total RNA purification using the Biomek® 2000 Laboratory Automation Workstation, the Vacuum Manifold (Beckman Coulter Part# 609670) and Collar (Beckman Coulter Part# 609597) are required. See the Automated Protocol, #EP003, for additional information on automated RNA purification.
