Quantify Endogenous Protein Expression in Real Time
HiBiT, an 11-amino-acid epitope tag, spontaneously interacts with LgBiT to generate the bright NanoBiT® luciferase. Introduction of HiBiT into target gene loci with CRISPR/Cas9 gene editing allows the study of protein under endogenous expression. Kinetically quantify HiBiT-tagged proteins in live cells from a single well using ViaScript™ LgBiT mRNA Delivery System and Nano-Glo® Extended Live Cell Substrates.
Benefits of Kinetic HiBiT Detection
- Observe effects missed by a single time point (e.g., hook effect).
- Accurately determine key compound characteristics, such as protein degradation/recovery rates, allowing therapeutic discovery, triage and optimization.
- Achieve more reproducible results by sampling the same well over time.
- Save time and reagents by not needing separate wells for each time point.
ViaScript™ LgBiT mRNA Delivery System offers faster and more uniform LgBiT expression to get more representative and precise live-cell kinetic HiBiT assay results.
