Leading the Pack: Canine PD-1/PD-L1 Blockade Bioassay
Canine immune checkpoint therapeutics are a recent development in companion animal medicine, building on the success of immune-checkpoint inhibitor treatment in humans. Similar to their human counterpart, the canine immune system relies on checkpoint molecules on immune cells to control an immune response, but this mechanism has also been exploited by canine tumors to avoid detection. Emerging studies in veterinary oncology are focused on discovering and developing canine-targeted antibodies for immune checkpoints, such as CTLA-4 and PD-1/PD-L1.
There are several challenges to developing canine-specific antibodies for immune checkpoints, such as low-efficacy and low-throughput screening measures due to lack of standardized reagents and protocols, leading to labor-intensive assays with high levels of variability.
The Canine PD-1/PD-L1 Blockade Bioassay is a two-cell-line system, a target cell that expresses canine PD-L1 and an effector reporter cell that expresses canine PD-1. The assay is a biologically relevant MoA-based and robust system that can be used to measure the potency and stability of biologics designed to block canine PD-1/PD-L1 interaction. This bioassay reduces data variability and provides a convenient luminescent readout.
Demonstrate Canine PD-1/PD-L1 Blockade
Principle of Canine PD-1/PD-L1 Blockade Bioassay. The Canine PD-1/PD-L1 Blockade Bioassay consists of two genetically engineered cell lines, the CHO-K1 target cells and the PD-1 reporter effector cell. When co-cultured, the PD-1/PD-L1 interaction inhibits TCR-mediated luminescence. When the PD-1/PD-L1 interaction is disrupted, TCR activation induces luminescence that can be detected by addition of Bio-Glo™ Reagent and quantitation with a luminometer.

Assay Performance Data
Canine PD-1 Effector Cells were co-cultured with Canine PD-L1 aAPC/CHO-K1 Cells in the presence of a titration of Anti-Canine PD-L1 Blocking Antibody. After 6 hours, Bio-Glo™ Reagent was added and plates were read on a GloMax® Discover Luminometer.
