RNasin®: The Most Trusted Name in RNase Inhibition

Promega RNasin® Ribonuclease Inhibitors are the gold standard for RNase protection. RNasin inhibitors feature in many publications and are the most widely used protection against RNases worldwide. Use RNasin® Ribonuclease Inhibitors for RNA isolation, RT-PCR, in vitro transcription or any situation where RNase  contamination is a concern.

RNasin® Plus RNase Inhibitor inhibits ribonucleases during reaction assembly. Heating of reactions to as high as 70°C for 15 minutes does not result in reappearance of ribonuclease activity, thus RNasin® Plus can be added prior to RT-PCR to offer enhanced protection of RNA during the reverse transcription reaction.

RNasin® Plus is Stable at Elevated Temperatures

RNasin® Plus RNase Inhibitor is a recombinant mammalian RNase inhibitor capable of inhibiting eukaryotic RNases, such as RNase A and RNase B, similar to the human version of the protein. Certain cysteine residues present in human placental RNase inhibitor have been implicated in the oxidation sensitivity of that enzyme (1). RNasin® Plus RNase Inhibitor does not contain these cysteine residues, which makes it more resistant to oxidative damage. RNasin® Plus RNase Inhibitor forms a stable complex with RNases, inactivating the RNase. This complex is stable at temperatures up to 70°C for at least 15 minutes (Figure 1). 

RNasin® Plus has been tested in RT-PCR and, like the human protein, is compatible with enzymes like AMV, M-MLV or ImProm-II™ Reverse Transcriptases as well as Taq and Tfl DNA Polymerases. RNasin® Plus has also been tested and found to be compatible with quantitative real-time RT-PCR in a TaqMan® Assay, and can even preserve templates from degradation in a biological extract as complex as a rat liver protein extract, demonstrating a broad spectrum of RNase inhibition (2).

One microgram of an RNA template was combined with 1μl of unheated RNasin® Plus (40 units), 2μl of unheated RNase A (20ng), 1μl of heated RNasin® Plus or 2μl heated RNase A as indicated.  In lane 7, 1μl of unheated RNasin® Plus and 2μl of unheated RNase A were combined before heating at 70°C for 15 minutes. Five microliters of the assembled reactions was then used as template for RT-PCR . See reference 2 for further experimental details.

Unit Definition

One unit is defined as the amount of RNasin® Ribonuclease Inhibitor required to inhibit the activity of 5ng of ribonuclease A by 50%. Activity is measured by the inhibition of hydrolysis of cytidine 2,3´-cyclic monophosphate by ribonuclease A.

20mM HEPES-KOH (pH 7.6 at 4°C), 50mM KCl, 8mM DTT and 50% (v/v) glycerol.
