The NanoBRET™ Target Engagement Intracellular Kinase Assay, Adherent Format technical manual describes the adherent (ADH) format of the NanoBRET™ TE Intracellular Kinase Assay. The ADH format is compatible with tissue culture-treated plates and allows measurement of TE in adhered cells, which may be more comparable to other cell-based phenotypic assays that utilize adhered cells. Compared to the legacy format that used nonbinding surface plates (NBS format), this new adaptation also creates a more efficient workflow. The new protocol in the ADH format allows the cells to be seeded into assay plates on day 1, enabling the user to proceed directly to tracer and test compound addition on day 2.This assay format is compatible with legacy tracers K-4 and K-5, as well as the new NanoBRET™ tracers (K-3, K-8, K-9, K-10 and K-11), and enables assay for over 250 unique kinases.
