HaloTag® Ligands
The HaloTag® technology portfolio offers ligands with a variety of functionalities, including fluorescent labels, affinity tags and solid-phase-attachment abilities. The ligands rapidly form covalent bonds under general physiological conditions; the bonds are highly specific and essentially irreversible. HaloTag® Fluorescent Ligands enable you to add the chloroalkane group, with which HaloTag® protein reacts, to any compound or surface with a compatible chemical group. The possible applications are endless.
Applications
- Pull Down Protein Complexes: The spacer and reactive linker of the HaloTag® PEG-Biotin Ligand provide ideal pull-down capabilities. Alternatively, pull down directly with the HaloLink™ Resin.
- Image Fixed Cells: The covalent bond is stable, allowing imaging of fixed cells and analysis of the labeled protein under stringent conditions.
- Introduce Novel Functionalities or Perform Sequential Labeling: The open architecture of the technology enables the use of different ligands for multiple applications.
- Design Only One Genetic Construct for Multiple Experiments: Obtain new functionality by using a different HaloTag® Ligand without having to design and clone a new expression construct.
- Analyze Labeled Fusion Proteins Using SDS-PAGE, Mass Spectrometry, etc.: The bound ligand is stable under denaturing conditions.

HaloTag® Technology Overview
HaloTag® Fluorescent Ligands for Cellular Imaging
We offer a variety of cell-permeant and cell-impermeant ligands for cellular imaging. Cell-permeant ligands readily cross the cell membrane and, therefore, can be used to label intracellular proteins. Cell-impermeant ligands, which cannot cross the cell membrane, can be used to label cell surface proteins.

Live cell labeling with impermeant HaloTag® Alexa Fluor® 488 and HaloTag® Alexa Fluor® 660 Ligands. U2OS cells stably expressing HaloTag® protein fused to the truncated beta 1 integrin receptor extracellular surface sequence or control U2OS cells expressing HaloTag® protein fused with three copies of a nuclear localization sequence were adhered to glass-bottom 96-well plates and labeled with Alexa Fluor® 488 or Alexa Fluor® 660 Ligand for 30 minutes at 37°C + CO2 in a cell culture incubator. Cell medium was replaced with a phenol-red-free medium. Cells were imaged with 488nm laser excitation for Alexa Fluor® 488 Ligand (Panel A) and 640nm laser excitation for Alexa Fluor® 660 Ligand (Panel B). In HaloTag-expressing cells, labeling was restricted to the membrane. Control U2OS cells expressing HaloTag in the nucleus showed no labeling. Images were collected using a Nikon AX/AXR confocal microscope with a 20X objective.
